In the chronic inflammatory microenvironment of tumors, lymphoid aggregates formed by the orderly assembly of innate and adaptive immune cells can occur within non-lymphoid tissues. Among these, mature structures possessing complete T and B cell zones and germinal centers are defined as tertiary lymphoid structures (TLSs). Numerous clinical studies have confirmed that the enrichment of TLSs in tumor tissues is closely associated with favorable prognosis in cancer patients and high response to immune checkpoint blockade (ICB) therapy, making them a highly promising novel therapeutic target in the field of cancer immunotherapy.
